Chinese Scientists Found the Important Genetic Mechanism of Bladder Cancer Stem Cells Origin

The cancer stem cell plays an important role in the occurrence, self-renewing, drug tolerance and transition of cancer cells. As the most common cancer in urinary system, the origin and genetic basis of Bladder Cancer Stem Cells are still unknown. The researchers from Chinese Academy of Science and Shenzhen University did the research to cancer stem cells from the aspect of genomics by single-cell sequencing technologies and carrying out the functional experiments.

The researchers carried out the unicellular MALBAC amplification and whole-exome sequencing to 59 cells which included Bladder Cancer Stem Cells, Bladder Cancer Non-Stem Cells, Bladder Epithelial Stem Cells and Bladder Epithelial Non-Stem Cells. Through phylogenetic analysis, they had found that Bladder Cancer Stem Cells start at Bladder Epithelial Stem Cells and Bladder Cancer Non-Stem Cells. Furthermore, they found that there are six genes which cannot be found in the bladder cancer by identifying the 21 key genes in Bladder Cancer Stem Cells mutation. The united mutation of ARID1A,GPRC5A and MLL2 can significantly enhance the capability that Bladder Cancer Non-Stem Cells convert into Bladder Cancer Stem Cells.

This research result describes the genomics overview of Bladder Cancer Stem Cells in details through the unicellular MALBAC amplification and whole-exome sequencing. It reveals the genetic basis of human Bladder Cancer Stem Cells origin and clarifies the self-renewal mechanism of which driver mutation in cancer cells regulate the Bladder Cancer Stem Cells. It is important for the prevention and treatment of human bladder cancer.
