China has built the world's largest maize mutant bank
Corn is not only the king of grain and feed, but also an important industrial raw material. The Chinese academy of agricultural sciences using chemical mutagenesis pollen successfully constructed B73 background nearly saturated mutant library, using explicit subgroups sequencing was carried out on 5894 mutant high-throughput sequencing and variation information mining, detected A total of 746607 potential cause changes in protein C - > T (G - > A) mutations, covering 36015 genes (91.59% coverage), corn cover 20 times the average gene.
On this basis, a sharing platform for maize mutants was built to provide mutant materials to researchers engaged in functional genomics of maize at home and abroad. Since the database of maize mutants was opened in 2017, more than 40 domestic and foreign research institutions and more than 140 research groups have distributed more than 8,000 copies of maize mutants.
Combined with forward genetics and reverse genetics, a new method of rapid and low-cost gene cloning, EcMutMap, was used to successively locate the regulatory genes such as middle vein of purple leaves, drought sensitivity and dwarfism. The mutant library and its matching gene cloning methods will greatly promote the development of functional genomics in maize in China. At the same time, more than 100 germplasm resources related to plant type, resistance, grain quality and straw quality were screened from the mutant library, providing rich genetic resources for the selection and breeding of new maize varieties.
